Title: Lysosomal System and Autophagy in Cancer
Abstract:The autophagolysosomal system, which combines autophagy and the lysosomal system, is a cellular quality control network that protects the health and homeostasis of cells by eliminating waste products like protein aggregates, damaged organelles, and pathogen invasion. As a result, the autophagolysosomal system plays a part in a wide range of pathophysiological conditions, including metabolic changes, cancer, neurological conditions, immune- and inflammation-related diseases, and more. The expression of numerous genes, including those encoding autophagolysosomal components, is controlled by the master transcriptional regulator TFEB, which also regulates the autophagolysosomal system. Reactive oxygen species (ROS) production and control are crucial components of the autophagolysosomal system’s physiopathological functions and may hold the key to synergistic therapeutic interventions. In this study, we reviewed our current understanding of the biology, physiopathology, and therapeutic potential of the autophagolysosomal system.
