Title: Understanding Pathogenic Mechanisms of Lysosomal Dysfunction with Lyso-IP
Abstract: All eukaryotic cells contain lysosomes, which are membrane-bound organelles that are ubiquitous. Beyond their well-known degradation role, lysosomes play a crucial role in preserving cellular homeostasis. A growing body of research has revealed that lysosomal dysfunction affects a variety of conditions, including common neurodegenerative diseases like Parkinson’s and Alzheimer’s, as well as the rare group of lysosomal storage diseases. In recent years, new technological developments have greatly improved our ability to quickly isolate lysosomes from cells. Lysosomal purification can now be completed in ten minutes thanks to the development of the Lyso-IP approach and related techniques. The enormous potential of this technique is demonstrated by the numerous studies using the Lyso-IP approach that have provided novel insights into the pathogenic mechanisms of lysosomal disorders, including Niemann-Pick type C disease. Our knowledge of lysosomal dysfunction in both uncommon and common neurodegeneration causes is likely to be greatly improved by future applications of rapid lysosomal isolation techniques.
