Title: Understanding Nuclear Receptors’ Functional Role in Esophageal Cancer
Abstract: One of the leading causes of cancer-related fatalities is esophageal cancer (EC), an aggressive and poorly understood disease. According to GLOBOCAN 2020, there will be 544,076 fatalities and 604,100 new cases worldwide. Despite numerous improvements in treatment methods, it has been reported that this cancer is one of the hardest to treat, and treatment targets still need to be established in order to increase patient survival. Nuclear receptors (NRs), a class of transcription factor, play a crucial role in many biological processes, including cellular differentiation, stress response, immunity, metabolism, lipid synthesis, and drug metabolism. They are also important regulators of a number of diseases, including cancer. The significance of NRs in tumor immunology and the established functions of multiple NRs in modulating proliferation, differentiation, and apoptosis have been proven in numerous studies. Few studies have shown the diagnostic and prognostic potential of NRs, despite the abundance of research on NRs and their effects on EC. Therefore, knowledge of the function of NRs and the various ways to target them in EC cells to prevent them from spreading cancer is still lacking. The discussion of various NRs’ agonists, antagonists, and responses to tumor progression in this review highlights the importance of NRs in EC. We also stress the ability of NRs to treat and prevent EC, as well as their potential to be a novel therapeutic target.
