Abstract: The large family of cell-surface receptors known as receptor tyrosine kinases (RTKs) is crucial for signal transduction pathways. There are multiple RTK subfamilies that can be used to classify the more than fifty human RTKs. RTKs play significant roles in the regulation of many cellular processes and mediate cellular signaling transduction. RTK signaling dysregulation is linked to a number of human diseases, including cancer. Numerous receptor tyrosine kinases have frequently been found to exhibit the proteolytic cleavage phenomenon. Richer insights have been gained as more and more information about proteolytic cleavage in RTKs has been uncovered. In order to better understand this phenomenon, we have compiled research on various facets of RTK cleavage, including its relationship to cancer. Proteolytic cleavage in various RTK members is also discussed in this review.
