Title: Two pairs of 7,70-Cyclolignan enantiomers from Perilla frutescens with anti-inflammatory properties
Abstract: Since about 1800 years ago, the medicinal plant Perilla frutescens (L.) Britt. (Labiatae) has been used extensively for the treatment of many different diseases. It has been established that P. frutescens extracts have potent anti-inflammatory properties. In this study, P. frutescens leaves were used to separate two pairs of 7,70-cyclolignan enantiomers with cyclobutanemoiety, (+)/()-perfrancin [(+)/()-1] and (+)/(()-magnosalin [(+)/(()-2]. The chiral separation of ()-1 and ()-2 was achieved in the current study, and their absolute configuration was identified. By lowering the secretion of pro-inflammatory factors (NO, TNF-, and IL-6) and the expression of pro-inflammatory mediators, compounds (+)-1 and ()-1 have notable anti-inflammatory effects (iNOS and COX-2). These results suggest that P. frutescens cyclolignans are potent anti-inflammatory compounds. The mechanisms underlying P. frutescens’ effects are only partially explained by the current study.
