Title: TRP Channels as Oxidative Stress and Aldehyde Sensors

Abstract: More than 50 channels make up the transient receptor potential (TRP) cation channel superfamily, which is important in physiological processes. Aldehydes have emerged as a biomolecule that can activate TRP channels, contributing to a cellular cascade that can result in disease pathophysiology. TRP channels are responsive to a variety of exogenous and endogenous biomolecules. In addition to endogenous aldehydes produced by lipid peroxidation, the body is also exposed to exogenous aldehydes from alcohol and tobacco products. Polyunsaturated fatty acids are converted to aldehydes like 4-hydroxynonenal as a result of lipid peroxidation brought on by inflammation or organ damage. Reactive aldehydes cause the release of pro-inflammatory mediators that drive the pathophysiology brought on by cellular injury, including inflammatory pain and organ reperfusion injury, via activation of TRP channels via aldehyde-induced protein adducts. Targeting the endogenous aldehyde metabolism may represent a novel treatment approach, as recent studies have shown how aldehyde dehydrogenase 2 protects against aldehyde toxicity through the clearance of toxic aldehydes. Targeting specific TRP channel regions can be an additional strategy to prevent aldehydes from triggering a cellular cascade. The role of aldehydes, TRP channels, and their interactions, as well as their function in pathological conditions and the various therapeutical treatment options, are all thoroughly covered in this review.
