Title: Treatment of Brain Metastasis from Breast Cancer Using C-Terminal Clostridium Perfringens Enterotoxin
Abstract: Claudin-4 is a member of the Claudin family of transmembrane tight junction (TJ) proteins that are present in almost all tissues. It forms epithelial and endothelial junctional complexes with adherens junctions and desmosomes. Claudin-4 is found in many different cell types, but the amount of expression varies depending on the type of cell. The expression of claudin proteins, which control cell proliferation and differentiation by binding cell-signaling ligands, is increased in a number of cancers. Changes in Claudin expression or distribution are therefore crucial to the pathology of cancer. Claudin-4 has a high degree of sequence homology with the high affinity receptor for clostridium perfringens enterotoxin (CPE), which was discovered through profiling the genetic expression of Claudin-4. Due to the high expression of Claudin-4 in breast cancer cells, CPE treatment has yielded promising results. CPE is cytolytic because it can create pores in cellular membranes. Given that Claudin-4 expression in the central nervous system (CNS) is low, the C-terminal fragment of CPE (c-CPE) offers a less toxic alternative for drug delivery into breast cancer cells, particularly metastatic tumors in the brain. As a result, the treatment of breast-brain metastatic tumors is made possible by c-CPE.
