Title: The Enteric Nervous System: Applications of Single-Cell Sequencing Technology
Abstract: Single-cell sequencing modalities are now widespread thanks to recent technological advancements and falling sequencing costs. These instruments enable single-cell analysis of RNA expression, DNA sequence, chromatin structure, and cell surface antigens. Multiple parameters can be measured simultaneously to resolve populations, including rare cells. This reveals the diversity of cells within organs and enables lineage reconstruction in developing tissues. The use of these techniques on the enteric nervous system has produced a wealth of information and biological understandings. We examine recent studies that used single-cell sequencing tools to study the developing enteric nervous system and the neural crest. These investigations have revealed a significant diversity of enteric neurons and glia, hypothesized neuronal specification paradigms, and identified signaling pathways that are active during development. We anticipate that multiome techniques that combine two or more transcriptomic, genomic, epigenetic, and proteomic data will become more popular as technology advances and become common in ENS research. These modalities may also play a role in diagnostic testing and personalized therapeutics.
