Title: structural and functional insights into the polymorphism of the -synuclein fiber
Abstract: A number of neurodegenerative diseases, such as Parkinson’s disease (PD), multiple system atrophy (MSA), dementia with Lewy bodies (DLB), Parkinson’s disease dementia (PDD), and even subsets of Alzheimer’s disease, are characterized by abnormal accumulation of aggregated -synuclein (-Syn). Lewy-body-like pathology in (AD). Similar to prion diseases, these synucleinopathies show variations in their clinical and pathological representations. Recent research indicates that -Syn self-assembles and polymerizes into a variety of conformational polymorphs both in vitro and in vivo. comparable to prions Upon their inoculation in animal models, these -Syn polymorphs, which are descended from the same precursor protein, may demonstrate strain-specific biochemical characteristics and the capacity to induce various pathological phenotypes. We discuss clinical and pathological variability in this review. in synucleinopathies and various -Syn fibril polymorphism features, including the presence of strains derived from the brain and high-resolution molecular structures. The review discusses the most recent developments in defining the structure-pathogenic relationship of -Syn and how various Synucleinopathies’ current clinical heterogeneity is a result of -Syn molecular polymorphs.
