Title: Prostate Cancer Extracellular Vesicle Proteome: Comparative Analysis of Mass Spectrometry Studies
Abstract: The promise of molecular diagnostics, based on discovery research, is to enhance prostate cancer screening procedures (PCa). The gathered data also begs the question of whether the urinary extracellular vesicles (uEV) proteome, particularly at the proteome level, has been thoroughly investigated. In actuality, plasma or serum have been the main targets of the majority of extracellular vesicle (EV) based biomarker studies. Therefore, the purpose of this study is to investigate potential methods for identifying urinary biomarkers, with a focus on the proteome of urine EVs. Reexamination of proteomics data stored in the PRIDE archive was done to identify potential PCa markers. There was some overlap among the markers suggested by various studies on prostate cancer, according to network analysis. A larger standardized cohort may be able to provide potential biomarkers that are able to completely characterize the heterogeneity of PCa, according to recent throughput improvements in mass spectrometry and the network analysis carried out in this study. Our analysis indicates that when compared to plasma, plasma EV, and voided urine proteome, PCa studies based on urinary EV proteome present higher protein coverage. This makes uEVs an appealing choice for protein biomarker studies, along with the fact that the prostate gland and urethra interact directly. In order to determine PCa specificity, urinary proteome-based PCa studies must also assess samples from cancers of the bladder and kidney.
