Title: Planar Chromatography and Chemometric Effect-Directed Profiling of Strawberry Varieties and Breeding Materials
Abstract: Because of their distinctive flavor, high concentration of vital nutrients, and bioactive compounds, strawberries are a significant fruit in the diet of Europe. In strawberries, the anthocyanins are known to be colorful phenolics. The anthocyanin profile as well as the antimicrobial and antioxidative activity profiles of 17 samples of six strawberry cultivars grown in Serbia, including the common varieties Alba, Asia, and Clery and promising breeding materials (11.29.11, 11.34.6, and 11.39.3), were determined. Gram-negative Aliivibrio fischeri was resistant to the antibacterial and antioxidant effects of all examined extracts. The volume and intensity of the responses were quite similar. Using HPTLC-DPPH scavenging assay, HPTLC-Alivibrio fischeri bioassay, and high-resolution mass spectrometry, it was discovered that the primary anthocyanin and prominent antioxidative and antimicrobial compound in strawberries is pelargonidin-3-O-glucoside (Pg-3-glc). Pg-3-glc quenches free radicals via sequential proton loss electron transfer mechanism in water and pentyl ethanoate, where the 5-OH group is the most reactive site for proton and hydrogen atom transfer, according to calculations using density functional theory at the M06-2X/6-31+G(d,p) level. Spectrophotometry was used to verify the results. Clery and 11.39.3 had the highest total phenolic content, but there were no statistically significant differences in antioxidant activity between the genotypes. Although the anthocyanin, antioxidative, and antimicrobial profile patterns of the strawberry genotypes were very similar, principal component analysis was able to successfully classify them.
