Title: Phase-Separated Prion-Like Proteins and Their Relation to Disease
Abstract: Numerous neurodegenerative illnesses, as well as some myopathies and cancers, are caused by abnormal protein folding. The presence of distinct prion and prion-like regions in some proteins can cause proteins to misfold. It has also been discovered that these prion and prion-like regions stimulate liquid-liquid phase separation. Thought to be a vital physiological process, liquid-liquid phase separation is also prone to error. Therefore, abnormal liquid-to-solid phase transitions may promote protein fibrillation and aggregation, which may result in pathological inclusions. In this article, we review prion diseases, prion-like proteins’ roles in phase separation, and potential treatment options for preventing abnormal phase transitions.
Keywords: prions; prion-like domains; amyloid; protein misfolding; liquid-liquid phase separation; aberrant phase transitions; chaperones
