Title: NLRP3 Inflammasome Targeting in Glaucoma
Abstract: The optic nerve head (ONH) is harmed in glaucoma, a group of optic neuropathies, causing vision loss in one or both eyes and retinal ganglion cell degeneration. One of the main aetiological risk factors for glaucoma is elevated intraocular pressure (IOP), which is also currently the only modifiable risk factor. However, 30–40% of glaucoma patients do not initially have an elevated IOP but still experience vision loss. Because of this, the pathophysiology of glaucoma is not fully understood, necessitating the creation of neuroprotective therapies that are independent of IOP in order to preserve vision. In particular, the NLRP3 inflammasome, a major driver of inflammation, has recently been linked to neuroinflammation, which has been shown to play a significant role in glaucoma. The NLRP3 inflammasome is expressed in the eye, and glaucoma pre-clinical studies have shown that it is activated. Processing of IL-1 takes place as a result of NLRP3 inflammasome activation. Axon degeneration and the death of retinal ganglion cells are thought to be caused by this pro-inflammatory cytokine, which is elevated in the blood of glaucoma patients (RGCs). The NLRP3 inflammasome is examined as a potential therapeutic target in this review, which also discusses glaucoma as an inflammatory disease. The NLRP3 inflammasome’s potential mechanism of action in glaucoma is presented.
