Title: Natural Flavonoids’ Promising Antiviral Activities Against SARS-CoV-2 Targets: A Systematic Review
Abstract: Over the past two years, the ongoing COVID-19 pandemic brought on by the SARS-CoV-2 coronavirus has become a major global public health concern. Despite the creation and use of numerous vaccines, challenges to universal immunity and the mutation of newer strains have shifted attention to the lack of effective therapeutic drugs for the disease. Given their success in silico, in vitro, in vivo, and more recently, clinical studies, flavonoids present themselves as a promising therapeutic intervention, similar to how they did with SARS-CoV, MERS-CoV, and other non-respiratory viruses. The analysis of the structure-activity relationships for the various key SARS-CoV-2 targets is the main focus of this review, which also examines data from in vitro studies looking at the effects of flavonoids on those targets. Over 69 flavonoids were examined for their effects on a variety of SARS-CoV-2 targets from the promising 3C-like protease (3CLpro) to the less studied nucleocapsid (N) protein, with the most promising compounds being derivatives of quercetin and myricetin, baicalein, baicalin, EGCG, and tannic acid. The therapeutic potential of extracts rich in flavonoids combined with synthetic drugs or other polyphenols is listed along with ongoing clinical studies. We also discuss promising in silico studies highlighting activities of flavonoids against SARS-CoV-2 and make predictions for the future of flavonoids against SARS-CoV-2.
