Abstract: The majority of the cell’s energy is produced by mitochondria, which also serve as communication hubs by directly and indirectly interacting with other organelles. Despite having a circular genome of its own, nuclear DNA encodes the majority of mitochondrial proteins. The mitochondria must communicate with the nucleus in order to adjust the metabolism or start a stress response in response to alterations in cell physiology. Retrograde signaling is the term used for this. Retrograde signaling encompasses a wide range of stimuli and pathways. It has already been established that mitochondrial dysfunction has negative effects on human health. Pathological deficits may also result from retrograde signaling disruption, whether it is directly linked to mitochondrial dysfunction or cellular environmental changes. Known signaling pathways between the mitochondria and the nucleus are discussed in this review along with potential direct contacts and pathological effects of a changed relationship.
