Title: Mechanisms, therapies, and biomarkers for NAFLD
Abstract: One of the leading causes of liver diseases worldwide is non-alcoholic fatty liver disease (NAFLD), which has recently been given the name metabolic-associated fatty liver disease (MAFLD). NAFLD is expanding at the same time that obesity is on the rise. There is no specific pharmacological treatment for NAFLD. The cause may be related to the multifactorial nature of NAFLD, the lack of adequate non-invasive biomarkers, inadequate imaging tools, and an incomplete understanding of the underlying mechanisms. In NAFLD, extra lipids build up in the liver, resulting in lipotoxicity, which can lead to hepatocellular carcinoma, metabolic-associated steatohepatitis (NASH), and liver fibrosis. This article discusses the mechanisms underlying NAFLD’s pathogenesis, current management strategies, and the function of sirtuins as potential therapeutic targets. Additionally, a summary of the current diagnostic techniques and the function of non-coding RNAs as developing diagnostic biomarkers is provided. The ability to detect the earliest NAFLD symptoms requires the availability of non-invasive biomarkers as well as precise, affordable non-invasive diagnosis tools. Clinical trials and the verification of novel therapeutic approaches will move more quickly as a result.
