Title: Location, Location, Location: HMGA1, Moonlighting Protein Function, and Cellular Real Estate!
Abstract: High levels of the gene encoding the chromatin remodeling protein High Mobility Group A1 (HMGA1) are associated with unfavorable clinical outcomes in a variety of cancers. Up until recently, it was believed that HMGA1 functioned as a nuclear protein that affected downstream signaling pathways and transcriptional regulation of genes to contribute to the development of cancer. However, the identification of an extracellular HMGA1-RAGE autocrine loop in invasive triple-negative breast cancer (TNBC) cell lines suggests that HMGA1 may act in multiple cellular locations as a “moonlighting protein” with various roles. Here, we review HMGA1’s function as a chromatin regulator in cancer and stem cells as well as its potential function as a secreted factor that promotes tumor growth. Previous research revealed that TNBC cell lines secrete HMGA1, which communicates with the RAGE receptor to promote phenotypes involved in tumor invasion and metastatic progression. Additionally, research on primary TNBC tumors indicates a link between HMGA1 secretion and distant metastasis in TNBC. Further investigation is required to confirm this function in other contexts given the therapeutic potential of extracellular protein targeting. In fact, interactions between nuclear and secreted HMGA1 may alter how we think about how tumors develop and open up new therapeutic possibilities for a variety of human cancers that overexpress HMGA1.
