Title: Lessons from the C. elegans Model on Sirtuins and Autophagy in Age-Associated Neurodegenerative Diseases

Abstract: Impaired protein clearance is one of the main factors contributing to the onset and progression of age-related neurodegenerative diseases. Therefore, as an organism ages, homeostasis is essential for preserving equilibrium within the cellular system. Any inconsistency in the protein clearance system causes unwanted proteins to accumulate, which has pathological effects such as neurodegeneration and related disability. There are many processes involved in controlling this phenomenon, but one crucial process that interferes with the protein degradation pathway and causes neurodegeneration is the inability to control the autophagic machinery. A group of proteins known as sirtuins is another significant and well-known factor that controls neurodegeneration. These histone deacetylases (HDACs) belong to the class III and are known to control a number of important biological functions, including DNA repair, genomic stability, and senescence. Additionally, it is known that these enzymes control neurodegeneration in an autophagy-dependent manner. The model system Caenorhabditis elegans has been successfully used in elucidating various functional outcomes related to crucial protein molecules, cell death pathways, and their association with aging. This is due to its genetic relevance and ease of studying disease related endpoints in neurodegeneration. This review highlights the knowledge gained using the C. elegans model system to highlight the critical role sirtuins and autophagy play in aging and neurodegeneration.
