Title: Is Cisplatin Therapy Always Compromised by Autophagy?
Abstract: For the most part used to treat solid tumors, cisplatin has long been a first-line chemotherapeutic drug in the fight against cancer. In studies involving cisplatin over the past two decades, autophagy has been almost always found to be activated in response to cancer treatments. Autophagy is now more widely acknowledged as a crucial element influencing tumor cell death and tumor chemoresistance. In this review and commentary, we outline four mechanisms of cisplatin resistance, discuss the variables that influence how autophagy functions in cisplatin-sensitive and resistant cells, and examine the contradictory effects of using autophagy inhibitors in conjunction with cisplatin. Our objective is to evaluate the potential for cisplatin and autophagy inhibitors to be used together in clinical settings.
