Title: Insights from Recent Structural Studies on Viral RNA-Dependent RNA Polymerases
Abstract: RNA viruses encode RNA-dependent RNA polymerases (RdRPs), a unique but adaptable class of nucleic acid polymerases for the replication and transcription of their genome. The RdRP has a structure resembling the fingers, palm, and thumb subdomains of a right hand held in a cup. Despite having a similar structural core, the RdRPs have vastly different mechanisms for RNA binding and polymerization. In light of recent structural analyses of RdRP complexes with various cofactors, RNA moieties, analogs, and inhibitors, the current review aims to explore these discrepancies.
