Title: Implications of Metabotropic Glutamate Receptors, a Family of Neuronal Receptors, in the Development and Progression of Cancer

Abstract:  The incidence of cancer is rising worldwide, and it is the second leading cause of death. Simply put, cancer is the unchecked growth of cells, and the etiology, biology, progression, prognosis, and treatment of cancer vary depending on the tissue of origin. Genetic components that are either tumor suppressors or oncogenes and can be either inherited or acquired are linked to the development of cancer. The MAPK and PI3K/AKT pathways, among others, are common signaling pathways where many of these genetic factors congregate. The metabotropic glutamate receptor (mGluR) family, an upstream protein that transmits extracellular signals into the cell and has been demonstrated to regulate numerous aspects of tumor development and progression, will be the main topic of this review. We investigate the role of this receptor family’s members in a number of cancers, such as T-cell cancers, pancreatic cancer, colorectal cancer, glioma, kidney cancer, melanoma, oral cancer, osteosarcoma, and pancreatic cancer. It’s interesting to note that, although most mGluRs function as oncogenes, they can also be categorized as tumor suppressors depending on the individual member. The extensive research done to understand the function of mGluRs in various cancers suggests that therapeutically targeting glutamatergic signaling may be a viable approach.
