Abstract: The absence of a mouse model of RhD immunization has made it difficult to study the immune response to the RhD antigen in the prevention of hemolytic disease in the fetus and newborn. However, this question has been reexamined in light of transgenic mice that express human HLA DRB1 1501 being able to react to purified RhD vaccination. In this study, we administered human RhD+ RBCs to mice transgenic for the human HLA DRB1 1501 as well as to a number of common inbred and outbred laboratory strains, including C57BL/6, DBA1/J, CFW(SW), CD1(ICR), and NSA. Our goal was to induce anti-RhD antibodies (CF-1). Additionally, putative extracellular immunogenic RhD peptide vaccinations were given to DRB1 1501 mice. After being immunized with RhD+ erythrocytes, DRB1 1501 mice produced antibodies that were reactive to erythrocytes. However, flow cytometry was unable to identify RhD-specific antibodies. Despite this, injection with Rh peptides caused antibodies reactive with RhD sequences, consistent with the presence of B cell repertoires capable of recognizing RhD, suggesting that DRB1 1501 mice were able to recognize immunogenic sequences of Rh. Conclusion: Immune responses to human RBCs expressing RhD are not directly seen, despite the possibility that HLA DRB1 1501 transgenic mice could react to immunogenic sequences within RhD.
