Title: Hepsin’s Clinical Relevance in Prostate Cancer and Underlying Signaling Pathways

Abstract: A type II transmembrane serine protease is encoded by the hepsin gene. Hepsin is overexpressed in prostate cancer, and hepsin dysregulation encourages cancer cell proliferation, migration, and metastasis both in vitro and in vivo, according to earlier studies. Hepsin expression levels were specifically higher in prostate cancer, according to the review that was incorporated into our work, and higher expression in metastatic tumors than in primary tumors was also noted. Additionally, elevated expression was linked to unfavorable outcomes for prostate cancer patients. Mechanistic analysis using in silico protein-protein interaction prediction revealed that eight other oncogenic proteins, whose expression was significantly correlated with hepsin expression in prostate cancer, interacted with hepsin. Hepsin’s oncogenic properties are primarily related to its proteolytic actions, which damage epithelial integrity and interact with other genes to regulate inflammatory, tyrosine-kinase, and cell-proliferation pathways. Furthermore, significant associations with prostate cancer metastasis were found with genomic hepsin amplifications rather than deletions or other alterations. Hepsin’s oncogenic behaviors are markedly reduced when it is specifically targeted with an inhibitor or antibody. Hepsin may thus serve as a new biomarker and therapeutic target for prostate cancer.
