Title: Genome stability and the role of transposable elements: implications for health and disease
Abstract: The majority of living things have a sizable portion of mobilizable DNA sequences in their genomes; these sequences are also known as transposons, transposable elements (TEs), or jumping genes. Despite being long believed to be of no biological importance, developments in DNA sequencing and analytical technologies have allowed for precise characterization of TEs and confirmed their pervasive presence across all forms of life. The biological significance of these discoveries has sparked heated debates. The evidence now supports the idea that TEs have a significant impact on a variety of biological aspects of organismal life. Transposable elements produce genetic variations in both active and passive ways, which significantly aids in the evolution of the genome. TEs primarily cause gene disruption and significant genomic alterations, such as inversions, deletions, and duplications, as a result of their inherent mobility within the genome. In addition to genomic instability, mounting evidence indicates that TEs have a wide range of physiologically significant roles, including the regulation of gene expression by cis-acting control elements and modulation of the transcriptome by epigenetic regulation. In this review, we discuss the most recent data showing how TEs affect genome stability as well as the underlying mechanisms, such as those created to lessen the negative effects of TEs on human health and genomic stability. We have also highlighted how TEs might be used therapeutically.
