Abstract: Renal and cardiovascular (CV) diseases are becoming more common worldwide and in the United States. Renal-related mortality is the eighth leading cause of death in the US, behind cardiovascular disease and cancer. Both diseases continue to exist despite effective treatments, necessitating further investigation into the pathophysiology of the diseases in order to create novel treatments and improve clinical outcomes for cardio-renal health. Age-related increases in CV and renal disease are observed, and there are sex differences in these conditions’ prevalence and course. These age and sex differences in cardio-renal health suggest that sex hormones may be significant regulators that need further investigation. G protein-coupled estrogen receptor 1 is a single example of a regulator (GPER1). GPER1 is expressed in many tissues, including the heart, vasculature, and kidney, and has been linked to estrogen signaling. Various experimental animal models have demonstrated the protective effects of GPER1 against renal and cardiovascular diseases. Numerous signaling pathways are involved in GPER1 actions, including interactions with endothelin-1 and aldosterone signaling, stimulation of nitric oxide production, and reduction of oxidative stress, inflammation, and immune infiltration. This review will go over the most recent research on GPER1 and cardio-renal health, especially in relation to aging. The development of novel therapeutics aimed at enhancing cardio-renal health and clinical outcomes in the elderly may be revealed by better understanding of GPER1-evoked mechanisms.
