Title: Fibulins’ Function in Embryonic Stage Development and Their Relation to a Range of Diseases
Abstract: Extracellular matrix (ECM) interactions between neighboring cells and the matrix itself play a crucial role in the evolution of early metazoans by supporting their structural and biochemical needs. ECM is crucial to the differentiation of tissues and the growth of organs in multicellular organisms. Fibulins are ECM glycoproteins that are linked to proteoglycan aggregates, elastic fibers, basement membranes, and fibronectin microfibrils in a variety of tissues. The role of fibulins in various developmental processes is revealed by their expression profile. such as elastic fibers, elastogenesis, organ development during the embryonic stage, tissue remodeling, preservation of the structural integrity of the basement membrane, and other cellular processes. In addition to this, fibulins play a role in the development of human diseases like chronic fibrotic disorders, cardiac diseases, congenital disorders, and cancer. Depending on the type of cell and cellular microenvironment in the body, different isoforms of fibulins exhibit a dual role of tumor-suppressive and tumor-promoting activities. In-depth understanding of their function in development and disease has been made possible by knockout animal models. The details of the structural and expression patterns, as well as the function of fibulins in embryonic development, are covered in the current review. with a focus on their contribution to the modulation of cancer diseases during disease progression.
