Title: Engineering Smooth Muscle to Understand Vascular Disease and Extracellular Matrix Remodeling
Abstract: The resident smooth muscle cells (SMCs) in blood vessel walls rely on specific mechanical and biochemical signals to carry out these functions, which are essential for controlling blood pressure and maintaining cardiovascular health. Any slight alteration in their environment prompts rapid phenotypic and secretory profile changes, which alter vessel wall structure and function and result in pathological conditions. It is crucial to comprehend how SMCs crosstalk with their surrounding extracellular matrix in order to effectively treat vascular diseases (ECM). Here, we review in vivo and conventional in vitro research on pathological vessel wall remodeling brought on by the SMC phenotype as well as the behavior of SMCs in response to important ECM properties. We then examine how modeling SMC responses to particular bodily stimuli is made possible by three-dimensional tissue engineering techniques. We also go over the effects of biomechanical forces and biochemical stimulation, like We are able to investigate disease mechanisms thanks to pulsatile fluid flow and factors secreted by other cell types. Overall, we suggest that high throughput experiments utilizing in vitro tissue engineering of human vascular smooth muscle can help to improve understanding of pertinent cardiovascular diseases, potentially resulting in future testing of therapeutics or treatments.
