Title: Elicitation Stimulates Lignan Production in In Vitro Cultures of Schisandra Rubriflora
Abstract: In the study, lignan accumulation in agitated and bioreactor (Plantform temporary immersion systems) microshoot cultures of female (F) and male (M) Schisandra rubriflora Rehd. et Wils. (Schisandraceae) lines was examined. Elicitation with chitosan (CH) (200 mg/L), yeast extract (YeE) (3000 mg/L), ethephon (ETH) (25 The 10th day of culture saw the addition of the elicitors. At 24 and 48 hours, as well as 4, 6, and 8 days after adding each elicitor, biomass samples were taken. Using the UHPLC-MS/MS technique, the 24 compounds from the lignans and neolignans dibenzocyclooctadiene, aryltetralin, dibenzylbutane, and tetrahydrofuran were determined both qualitatively and quantitatively in biomass extracts. The highest total lignan contents [mg/100 g DW] were found in CH-95.00 (F, day 6) and 323.30 (M, 48 h); YeE 104.30 (F, day 8); ETH 124.50 (F, 48 h); and MeJA 89.70 (F, 48 h) and 368.50 (M, day 4). (M, 24 h). The highest total lignan content (153.20 mg/100 g DW) was found in the biomass extracts of M cultures grown in bioreactors after MeJA elicitation. The maximum total lignan contents were 3.29 and 1.13 times higher in the biomass extracts from the agitated and bioreactor cultures than in the nonelicited cultures, respectively. The ingenuity of the research is highlighted by our limited understanding of S. rubriflora’s chemical makeup and the paucity of studies in plant biotechnology.
