Title: Dysfunction of the endothelium has been implicated in the development of abdominal aortic aneurysms.
Abstract: A common and potentially fatal vascular condition is known as an abdominal aortic aneurysm (AAA), which is defined as a focal dilation of the abdominal aorta beyond 50% of its normal diameter. Uncertainty exists regarding the molecular and cellular mechanisms underlying AAA pathogenesis. By controlling vascular tone and preserving a local environment that is anti-inflammatory and anti-thrombotic, healthy endothelial cells (ECs) play a crucial role in preserving vascular homeostasis. A growing body of research suggests that endothelial dysfunction, which contributes to oxidative stress and inflammation in the aging arterial wall, is an early pathologic event in AAA formation. Recent research using single-cell RNA sequencing identified diverse EC sub-populations in aortic aneurysm tissue based on their transcriptional profiles. Recent research on the effects of biomechanical stress on EC in AAA, the role of endothelial nitric oxide synthase (eNOS) uncoupling in AAA, and EC heterogeneity in AAA are all summarized in this review. Clinical evidence of endothelial dysfunction in AAA is also covered. These investigations advance our knowledge of AAA pathogenesis and may ultimately result in the development of EC-targeted therapeutics for the treatment or avertance of this fatal condition.
