Title: Biomaterials-Based Approaches for the Treatment of Volumetric Muscle Loss in Skeletal Muscle Tissue Engineering

Abstract: Every year, millions of Americans experience skeletal muscle injuries that can lead to volumetric muscle loss (VML), a condition in which severe musculoskeletal damage and tissue loss leave patients with long-term functional deficits. Skeletal muscle is capable of endogenous regeneration in cases of minor injuries thanks to the activation of local satellite cells (SCs). However, in VML injuries, which eliminate native biophysical and biochemical cues and impair the injured tissue’s capacity to direct regeneration, this is significantly diminished. Autologous tissue transfer is the current clinical treatment for VML, but graft failure and scar tissue development leave patients with restricted functional improvement A promising method for treating VML injuries involves the use of instructive biomaterial scaffolds for tissue engineering. Here, we discuss how to strategically incorporate biophysical and biochemical cues into current scaffold designs to help treat these preclinical VML injuries and restore function. The three primary biomaterial-based approaches to treating VML injuries—acellular scaffolds, cell-delivery scaffolds, and in vitro tissue engineered constructs—are also covered, along with their advantages and disadvantages. In order to improve the functional regeneration of skeletal muscle after VML injuries, we examine a number of novel approaches to improving the design of the next generation of engineered scaffolds.
