Title: Biogenic gold nanoparticles are effective against Pseudomonas aeruginosa as antibacterial and antibiofilm nano-antibiotics.

Abstract: There are numerous uses for environmentally friendly nanoparticles made from plants, including in medicine. Once formed, biofilm—a defense mechanism for pathogenic microorganisms—makes it challenging for antibiotics to eradicate the pathogen. Using the stem of the Ayurvedic herb Tinospora cordifolia, we created gold nanoparticles (AuNPs) and investigated how they affected the Pseudomonas aeruginosa PAO1 biofilm. Techniques like scanning electron microscopy (SEM), Fourier-transform infrared (FTIR) spectroscopy, energy-dispersive X-ray diffraction, and transmission electron microscopy were used to characterize the synthesized AuNPs. The AuNPs had spherical shapes and were 16.1 nm in size on average. Additionally, the crystal violet assay and SEM revealed that P. aeruginosa’s ability to form biofilms was significantly impacted by the subminimum inhibitory concentrations (MICs) of AuNPs (50, 100, and 150 g/mL), which decreased the number of biofilm-forming cells with increasing AuNP concentration. Confocal laser scanning microscopy (CLSM), which revealed irregularities in the structure of the biofilm at the sub-MIC of AuNPs, further supported this. Additionally, the internalization of the nanoparticles was evident during the interaction of AuNPs with PAO1 at the highest sub-MIC (150 g/mL), which likely affected PAO1’s propensity to colonize on the surface of the nanoparticles. According to this research, P. aeruginosa-caused biofilm-related infections can be effectively treated with green-synthesised AuNPs.
