Title: Bioactive Scaffolds for Bone Regeneration Integrated with Liposomal or Extracellular Vesicles
Abstract: An increasing number of people are experiencing musculoskeletal health issues that call for therapeutic intervention at defect sites as a result of population aging and rising life expectancy. A promising alternative to traditional bone grafts is bone tissue engineering (BTE). Clinical complications can occur during autograft or allograft-based treatments. The creation of ideal BTE scaffolds that can incorporate bioactive molecules promoting robust bone repair has come a long way. In this article, we examine bioactive scaffolds designed for local bone regeneration. therapy, in particular by adding extracellular vesicles or synthetic liposomal vesicles to the scaffolds. An excellent drug delivery system, liposomes allow for the sustained release of the bioactive molecules they contain. Extracellular vesicles are emerging as a cutting-edge replacement for synthetic nanoparticles and a cutting-edge cell-free therapy for bone regeneration due to their inherent ability to carry bioactive molecules. We go over recent developments in the application of synthetic liposomes and Extracellular vesicles as bioactive materials combined with scaffolds, highlighting significant difficulties and possibilities for their use in bone regeneration. Our focus is on methods for integrating vesicles into different biomaterial scaffolds, and we also present the most recent developments in achieving sustained release of bioactive molecules from the vesicle-loaded scaffolds at the site of the bone defect.
