Title: Based on Solid-Phase Microextraction and Ultrahigh-Performance Liquid Chromatography-Tandem Mass Spectrometry, In Vivo Detection of Tetrodotoxin in Takifugu obscurus
Abstract: Although pufferfish is tasty and nutritious, the tetrodotoxin (TTX) that could be present in it poses a serious safety risk. The detection of TTX in pufferfish requires the use of accurate scientific techniques, but most of the currently available approaches have sample lethality and complicated pre-treatment procedures. Due to its benefits like no solvent requirement, straightforward operation, and quick detection speed, solid-phase microextraction (SPME) technology can be used for in vivo detection. The GO-PAN@PNE SPME fibers were created in this study using a dipping technique, and the TTX aqueous and spiked fish were used to confirm their extraction effect. The limit of detection of TTX in pufferfish was 32 ngg-1, and the limit of quantitation was 150 ngg-1, which can meet the detection requirements of pufferfish for safe consumption. The established method has good reproducibility. With the help of this technique, it was possible to identify Takifugu obscurus in vivo, as well as the amount of TTX present in the pufferfish muscle. The in vivo detection of TTX in pufferfish can be achieved relatively quickly and easily using the detection method developed in this study, which can offer theoretical support for an increase in the pufferfish’s level of food safety.
