Abstract: Epitopes in wheat gluten can cause celiac disease (CD). The only accepted treatment for CD is a strict gluten-free diet for the rest of one’s life. Very low-gluten wheat, however, might offer a substitute for CD treatment. Wheat that is safe for celiacs has not been produced using conventional plant breeding techniques. In some ways, the development of safer wheat varieties has been successful thanks to RNA interference technology. When it comes to implementation, these varieties face numerous difficulties. The versatile gene-editing tool CRISPR/Cas9 (Clustered Regularly Interspaced Short Palindromic Repeats-associated Nuclease 9) has the capacity to modify the genes encoding the immunogenicity of gluten. Only a few studies have used CRISPR/Cas9 to alter the wheat genome thus far. In order to investigate the current status of the CRISPR/Cas9 system to create a low-immunogenic wheat variety, we reviewed the published literature that used CRISPR/Cas9 in wheat genome editing. We discovered that the CRISPR/Cas9 system has been enhanced continuously in recent years to edit the challenging hexaploid wheat genome. Although some wheat varieties with reduced immunogenicity have been reported, CRISPR/Cas9 has not yet been used extensively to edit the wheat genome. We come to the conclusion that more research is necessary to effectively use the CRISPR/Cas9 gene-editing system for the creation of a wheat variety that is safe for people with celiac disease and to establish it as a “tool to celiac safe wheat.”
