Title: An Important Cytokine in Pathologies and Cancer: Leukemia Inhibitory Factor
Abstract: Leukemia Inhibitory Factor (LIF), an IL-6 cytokine family member, is expressed in almost all body tissue types. Although myeloid leukemia cell differentiation was the reason LIF was given its name, research on LIF in other diseases and solid tumor types has shown that it has the potential to affect a wide range of pathologies. Investigating the functions of LIF in healthy physiology and pathologies unrelated to cancer can provide crucial insights into how this protein may be dysregulated within cancers and the potential consequences of this dysregulation. LIF expression has been connected to cancer hallmarks like proliferation, metastasis, and chemoresistance as well as general patient survival across a range of cancer types. The mechanisms underlying these LIF effects are unclear and can vary between various tissue types. In fact, studies have shown that LIF can have anti-neoplastic effects in some solid tumors while potentially promoting malignancy progression in others. This review will summarize the current understanding of how LIF expression affects cellular function and dysfunction in order to identify potential adjuvant treatment side effects as well as new treatment options for cancer patients.
