Title: Acetaldehyde’s anticoagulant effect on APTT is affected differently by aromatic amines.
Abstract: Amphetamine, procaine, procainamide, DOPA, isoproterenol, and atenolol have all been studied for their pharmacological effects on activated partial thromboplastin time in the presence and absence of acetaldehyde. Amphetamine and isoproterenol have a procoagulant effect on activated partial thromboplastin time in the absence of acetaldehyde, whereas atenolol and procaine have an anticoagulant effect on this same measure. The activated partial thromboplastin time is not changed by DOPA or procainamide. Procaine and acetaldehyde premixtures have an additive anticoagulant effect on activated partial thromboplastin time, indicating that these compounds act independently on the clotting factors. The anticoagulant effect of acetaldehyde is detoxicated upon activated partial thromboplastin time in premixtures of amphetamine and acetaldehyde, as well as atenolol and acetaldehyde. When procainamide and acetaldehyde are mixed for 20 minutes at room temperature, a comparable statistically significant reduction in the activated partial thromboplastin time is observed. When combined with acetaldehyde, DOPA and isoproterenol do not alter the activated partial thromboplastin time in comparison to acetaldehyde alone. The presence of amino, hydroxyl, or amide groups in atenolol, procaine, and amphetamine suggests a selective interaction with acetaldehyde to result in detoxication of the acetaldehyde.
