Abstract: Over the past few years, tremendous strides have been made in our understanding of the molecular mechanisms underlying chronic diseases, but they are still the leading cause of death in the world. Most of the approved drugs used to prevent and treat these diseases focus on just one gene, protein, or pathway, have a history of serious side effects, and perform less well than expected. As a result, there is great potential for the development of better therapeutics that surpass those that already exist. Numerous deadly diseases can be prevented and treated with natural compounds. Oroxylin A (OA), a flavonoid with unique pharmacological characteristics, is derived from the plants Oroxylum indicum, Scutellaria baicalensis, and S. lateriflora. NF-B, MAPK, ERK1/2, Wnt/-catenin, PTEN/PI3K/Akt, and signaling molecules like TNF-, TGF-, MMPs, VEGF, interleukins, Bcl-2, caspases, HIF-1, EMT proteins, Nrf-2, etc. are important signaling pathways that OA modulates. These pathways and signaling molecules are crucial in the molecular mechanism of chronic diseases. The potential for this flavonoid to be anti-inflammatory, anti-bacterial, anti-viral, and anti-cancer is overwhelmingly supported by evidence, which makes it an intriguing substance for research. Numerous preclinical and clinical studies also demonstrated the promising effects of OA against conditions like rheumatoid arthritis, osteoarthritis, cancer, cardiovascular disease, inflammation, and neurological disorders. As a result, the current review concentrates on outlining OA’s role in battling various chronic diseases and emphasizing the inherent molecular mechanisms of its action.
