Title: A detailed analysis of 3D structure holdings at the Protein Data Bank and its use by scientists, teachers, and students worldwide

Abstract: Worldwide support for structural biologists and Protein Data Bank (PDB) data users is provided by the Research Collaboratory for Structural Bioinformatics Protein Data Bank (RCSB PDB), which is funded by the National Science Foundation, National Institutes of Health, and Department of Energy of the United States. The RCSB PDB is the US data center for the global PDB archive, which houses experimentally determined three-dimensional (3D) structure data for biological macromolecules. The RCSB PDB is a founding member of the Worldwide Protein Data Bank (wwPDB) partnership. The security of PDB data and weekly updating of the archive are additional responsibilities of RCSB PDB in its capacity as the wwPDB’s designated Archive Keeper. Tens of thousands of data depositors use the RCSB PDB each year, working on all permanently inhabited continents using macromolecular crystallography, nuclear magnetic resonance spectroscopy, electron microscopy, and micro-electron diffraction. Many millions of PDB data users worldwide have access to RCSB PDB’s PDB data for free and with no usage restrictions through its research-focused RCSB.org web portal. Through its web portal for outreach and education, PDB101.RCSB.org, it also offers educators, students, and the general public an introduction to the PDB and related training materials. This review article provides a thorough accounting of the PDB archive’s holdings and its use by researchers, educators, and students across the globe. It also discusses the PDB’s growth and the evolution of experimental methods for structure determination as seen through the prism of the PDB archive.
