Title: A Comprehensive Review of the Literature on Drug Repurposing for Glioblastoma and Current Advances in Drug Delivery
Abstract: The most frequent primary malignant brain tumor in adults, glioblastoma (GBM), has a very poor prognosis. To overcome the inherent and acquired resistance of GBM to current therapies, it is urgently necessary to develop effective therapeutics. Novel anti-neoplastic drug development can take a long time and be expensive, from the lab to the patient’s bedside. Drug repurposing could assist in overcoming that difficulty. Numerous medications that have already received clinical approval for the treatment of other illnesses have been discovered to target signaling pathways linked to GBM and are currently being repurposed for the treatment of GBM. While many of these medications are still in the pre-clinical testing stage, others are currently undergoing clinical trials. Recent studies have also looked into whether repurposed drugs that target these pathways can be used to prevent tumor recurrence since GBM stem cells (GSCs) have been found to be a major cause of tumor recurrence after surgery. Even though a number of repurposed medications have demonstrated significant efficacy against GBM cell lines, the blood-brain barrier (BBB) can prevent many of these medications from reaching therapeutic intratumoral concentrations. Localized intracranial delivery could reduce toxicity and side effects while achieving therapeutic drug concentration at the site of tumor resection. These tactics can be taken into account when repurposing medications for GBM.
