Title: Natural Ergot Alkaloids for Novel Nanoparticle-Based Delivery Systems in Ocular Pharmacotherapy
Abstract: Because of their antibacterial, antiproliferative, and antioxidant effects, ergot alkaloids are thought to possess a variety of pharmacological properties. The majority of ergot alkaloids exhibit high toxicological risk and may even be fatal to humans and animals, despite the fact that they are known for their biomedical applications (such as the treatment of glaucoma). Their pharmacological profile is a result of the structural resemblance between lysergic acid-derived substances and the neurotransmitters noradrenaline, dopamine, and serotonin. Improved delivery systems were suggested to decrease their toxicological risk while increasing their bioavailability. In order to increase the therapeutic efficacy of these medications for the treatment of glaucoma, this review suggests the development of lipid and polymeric nanoparticles for topical administration of these medications. It also discusses the safety aspects of using ergot alkaloids in ocular pharmacology.
