Title: At the Line Between Inside and Outside, Loricrin
Abstract: The cell-death program’s specialized mode of cornification is only available to terrestrial amniotes. Loricrin (LOR), according to recent studies, is a crucial cornification effector. The keratin-associated protein LOR promotes the maturation of the epidermal structure by organizing covalent cross-linkages, giving the epidermis protection against oxidative injuries, as the name’s connotation (“lorica” means “armor” in Latin). We aim to classify ichthyosiform dermatoses according to their function rather than clinical manifestations by reviewing cornification mechanisms. As LOR and NRF2 coordinate the epidermis-intrinsic xenobiotic metabolism, we also discussed recent mechanistic findings about the role of the Kelch-like erythroid cell protein with the cap “n” collar homology-associated protein 1/nuclear factor erythroid 2-related factor 2 (NRF2) signaling pathway in skin health and diseases. The theoretical model of cross-talk between keratinocytes and epidermal resident leukocytes is then improved, and a LOR immunomodulatory function is analyzed.
