Title: A Potential Target for the Treatment of Psychiatric Disorders Is PPAR Signaling
Abstract: Peroxisome proliferator-activator receptors (PPARs) control inflammatory processes, modulate a number of brain functions, and regulate lipid and glucose metabolism. There are three distinct PPAR isoforms—PPAR, PPAR/, and PPAR—that are expressed in various tissues and cell types. In the sections that follow, we will concentrate on PPAR’s role in neurodevelopment and neuroinflammation, as well as its involvement in the pathophysiology of neuropsychiatric and neurodegenerative disorders. PPAR is localized in neuronal circuits that regulate emotion and the stress response. Mechanisms underlying behavioral regulation include a variety of downstream pathways that are modulated by PPAR activation, such as glutamatergic neurotransmission, upregulation of brain-derived neurotrophic factor, and neurosteroidogenic effects. Effects of PPAR in depression, anhedonia, and autism spectrum disorder are most likely influenced by modulation of dopamine neuronal firing in the ventral tegmental area (ASD). Future clinical trials should evaluate the effectiveness of PPAR agonists in the treatment of mood and neurodevelopmental disorders, such as depression, schizophrenia, and ASD, based on strong preclinical evidence and the preliminary findings of clinical studies.
