Title: Past and Present Cancer Target Ribonucleotide Reductase Inhibitors
Abstract: All living things contain the multi-subunit enzyme ribonucleotide reductase (RR), which catalyzes the rate-limiting step in the synthesis of deoxyribonucleoside diphosphates, which is the conversion of ribonucleoside diphosphates to ribonucleoside diphosphates. Human RR (hRR), which is highly expressed during cell replication, has long been regarded as a promising drug target for a variety of proliferative diseases, including cancer. Although there are many excellent reviews of the structure, function, and clinical significance of hRR, more novel approaches to inhibiting hRR have emerged in recent years, necessitating a discussion of the current inhibitors and tactics to target this enzyme. The mechanisms and clinical uses of well-known nucleoside analog inhibitors of the hRRM1 (large catalytic subunit), such as gemcitabine and clofarabine, as well as inhibitors of the hRRM2 (free radical housing small subunit), such as triapine and hydroxyurea, are covered in this review. We also go over fresh methods for RR targeting and the identification of fresh classes of hRR inhibitors.
