Title: Peptidomimetics and Their Uses in the Development of Opioid Peptide Drugs
Abstract: Opioid peptides have a variety of benefits, but their therapeutic applications have been constrained by their main drawbacks in metabolic stability, blood-brain barrier permeability, and bioavailability. As a result, in-depth research has concentrated on solving the issues and maximizing the therapeutic potential. Many peptide-based drugs are currently on the market as a result of new synthetic techniques for enhancing metabolism and alternative routes of administration. The history and function of natural opioid peptides are briefly discussed in this tutorial review, along with the most important discoveries regarding their structure-activity relationships for opioid receptors. The pharmacological and structural aspects of opioid peptidomimetics used to create therapeutic candidates for the treatment of pain are covered in detail. The current state of various mimetic tools and the successful applications are the main points of emphasis, and they are summarized in tables and figures.
