Abstract: Alzheimer’s disease (AD) is a neurodegenerative condition that progresses over time and has a multifactorial onset. Early pathological signs of neuroinflammation, neurofibrillary tau and amyloid pathologies include a deficit in neurogenesis and synaptic plasticity. Other characteristics of AD include disruption of the brain microenvironment and an imbalance of neurotrophic factors, with an increase in FGF-2 level and a decrease in BDNF and NT-4 in the hippocampus, frontal cortex, and parietal cortex. The differentiation, maturation, and survival of neurons depend heavily on neurotrophic factors. There have been several attempts to use neurotrophic factors to treat AD, but these studies have been stopped due to the BBB’s impermeability, short half-lives, and negative side effects. In the current review, we primarily concentrate on the key etiopathology characteristics of AD and the use of P021, a small neurotrophic and neurogenic peptide mimic that was discovered in our lab and was found to be effective in overcoming the challenges associated with the administration of the full neurotrophic factor proteins. In this article, we discuss pre-clinical research on P021 and its potential as a treatment for AD and other neurodegenerative diseases. Our research is constrained because it only examines P021 and the pertinent literature. A more thorough investigation is needed to review studies on various therapeutic modalities and potential medications that are being developed in the field of AD.
