Title: Beyond Mineralization: The Physiological and Pathological Role of Tissue Nonspecific Alkaline Phosphatase
Abstract: The important enzyme Tissue-nonspecific Alkaline Phosphatase (TNAP) is in charge of the mineralization of skeletal tissue. It performs essential physiological functions, including extra-skeletal functions like neuronal development, and is involved in the dephosphorylation of a variety of physiological substrates. LPS detoxification, anti-inflammatory activity, control of bile pH, and preservation of the blood-brain barrier are all functions of bile (BBB). Additionally, ectopic pathological calcification of soft tissues, particularly the vasculature, is linked to TNAP. Despite being the essential enzyme for mineralization It is a logical clinical goal to lessen vascular calcification of skeletal and dental tissues. To inhibit its activity and halt the mineralization of soft tissue, numerous tools and studies have been created. Prior to treatments that target TNAP, we should not disregard its other physiological functions. Therefore, a deeper comprehension of the mechanisms mediated by TNAP is required to reduce targeted effects and improve a variety of pathological scenarios. This review includes have talked about how minerals are formed and how TNAP serves other purposes besides just mineralizing hard tissues.
