Title: An Accurate Method to Demystify Microbial Metabolism for Biochemical Production: 13C-Metabolic Flux Analysis
Abstract: Because it is environmentally friendly, sustainable, and independent of fossil fuels, metabolic engineering of various industrial microorganisms to produce chemicals, fuels, and medications has gained interest. resources that are not renewed. Only a few metabolic engineering initiatives, however, have been successful in producing a satisfactory yield, titer, or productivity of the target chemicals for industrial commercialization because microbial metabolism is so complex. The 13C Metabolic Flux Analysis (13C-MFA) has been continuously developed and widely used to rigorously investigate cell metabolism and quantify the carbon flux distribution in key metabolic pathways in order to meet this challenge. To identify the main problems with microbe-based chemical production, numerous 13C-MFA studies have been carried out in academic labs and the biotechnology sector over the last ten years. The development of the appropriate metabolic engineering strategies for enhancing biochemical production has been guided by insightful information about the metabolic rewiring. We will introduce the fundamentals of 13C-MFA in this review and show how it has been used in conjunction with metabolic engineering to pinpoint and address rate-limiting steps in the biochemical production of a variety of host microorganisms.
