Title: A Bottom-Up Approach to Understanding Human Neurodevelopment Using Brain Organoids
Abstract: Recent advances in three-dimensional tissue culture have made it possible to study the fundamentals of neurodevelopment and morphogenesis using brain organoids. Importantly, since human stem cells can be used to create brain organoids, they provide a model system for studying early human brain development and human-specific disorders. However, there are still a lot of significant differences between in vivo development and in vitro systems. This is partly a result of the difficulty in creating an appropriate cultural platform that will support proper development. In this review, we examine the analogies and contrasts between the organoid systems of the human brain and those of the embryo. The use of organoids to model neurodevelopmental diseases is then discussed. The challenges in organoid systems are then discussed, along with complementary bioengineering methods for solving them.
