Title: Hydrogel Models for Investigating Pancreatic Cancer Cell Fate with Stiffness Gradients

Abstract: The most prevalent form of pancreatic cancer is pancreatic ductal adenocarcinoma (PDAC), and patient survival rates have only slightly increased in recent years. Due to the presence of a dense and hypovascularized fibrotic tissue made up of stromal cells and extracellular matrices, PDAC is extremely aggressive and resistant to chemotherapy. A more diverse microenvironment is created by stromal cells’ increased matrix deposition and crosslinking, which promote PDAC advancement. Several hydrogel-based in vitro tumor models have been created over the last ten years to mimic and recapitulate different aspects of the tumor microenvironment in PDAC. Hydrogel chemistry and engineering developments should offer a forum for learning new information about how matrix characteristics control the expansion, invasion, and drug resistance of PDAC cells. These engineered hydrogels are perfect for figuring out how differences in matrix properties, such as durotaxis—the directional migration of cells in response to a stiffness gradient—contribute to the propensity of cancer cells to spread. The different hydrogel-based in vitro tumor models are covered in this review, along with techniques for producing gradient stiffness to study migration and other cancer cell fate processes in PDAC.
