Title: Human Mesenchymal Stem Cell Bioprocessing: Microcarrier-Based Bioreactors Versus Planar Culture
Abstract: Human mesenchymal stem cells (hMSCs) have shown great promise as treatments for a wide range of illnesses. Allogeneic hMSC therapies are particularly appealing options and methodologies to improve their scaling and manifolding because of their immunoprivileged status. Manufacturing is required. In automated closed systems, microcarrier-based bioreactor systems produce more hMSCs per volume than traditional planar cultures. However, the successful transition from planar culture to microcarriers requires more advanced bioprocesses. The main steps in the planar culture to microcarrier hMSC manufacturing scheme are outlined in this article, including seed train, inoculation, expansion, and harvest. Important bioreactor parameters are also covered, including temperature, pH, dissolved oxygen (DO), mixing, feeding techniques, and cell counting methods.
